Domestication of Haliotis tuberculata has only recently begun. During the process, we expect that behavioural and physiological traits may evolve to become more adapted to their captive environment. These modifications may result from intentional selection of production traits or unconscious and unintentional selection due to conditions experienced in the farm environment. Development of a polymerase chain reaction for the detection of abalone herpesvirus infection based on the DNA polymerase gene ArchiMer

A 5781-base pair (bp) fragment of genomic DNA from the Taiwanese abalone herpesvirus was obtained and showed 99% (5767/5779) homology in the nucleotide sequence and 99% (1923/1926) in the amino acid sequence with the DNA polymerase gene of the abalone herpesvirus strain Victoria/AUS/2007. Homology of the amino acid sequence with the DNA polymerase of ostreid herpesvirus 1 was 30% (563/1856). In this study, a PCR-based procedure for detecting herpesvirus infection of abalone, Haliotis diversicolor supertexta, in Taiwan was developed. Immune priming and portal of entry effectors improve response to vibrio infection in a resistant population of the European abalone ArchiMer

Since 1997, populations of the European abalone Haliotis tuberculata suffer mass mortalities attributed to the bacterium Vibrio harveyi. These mortalities occur at the spawning season, when the abalone immune system is depressed, and when temperatures exceed 17 °C, leading to favorable conditions for V. harveyi proliferation. In order to identify mechanisms of disease resistance, experimental successive infections were carried out on two geographically distinct populations: one that has suffered recurrent mortalities (Saint-Malo) and one that has not been impacted by the disease (Molène). The commercial culture of Haliotis tuberculata has recently started in Europe. As abalone is herbivorous, the use of local collected algae as feed may appear advantageous. The nutritional value of eight monospecific seaweed diets was studied using Palmaria palmata (Rhodophyta), filamentous algae, mainly Gracilaria sp. Abalone aquaculture only recently developed in western Europe, but already it has to face numerous pathologies. Since 1998, the natural and farm stocks of Haliotis tuberculata suffer regularly high mortalities, mainly due to the pathogenic bacterium, Vibrio harveyi. The vibriosis of the European abalone, in particular its macroscopic signs, and the conditions necessary for its development have been studied in detail by field and laboratory experiments. Les mortalités d'ormeaux en Bretagne nord en 1998: synthèse des observations et analyses ArchiMer

Des mortalités anormales d'ormeaux ont été signalées en Bretagne nord (départements d'Ille et Vilaine et Côtes d'Armor) à partir du mois d'août 1998 et porté atteinte à la pêcherie professionnelle de cette zone. Des mortalités de même type avaient été signalées en Bretagne Sud, l'année précédente. Les mortalités de 1998 ont épargné le secteur plus à l'ouest (le Finistère) et plus à l'est (la Normandie). Une estimation précise n'a pas été entreprise, mais les pertes signalées par endroits dépassent 50% du stock. Les ormeaux atteints, de toutes tailles, ne présentent pas de symptôme particulier. In the context of an increasing demand and a declining supply from fisheries, worldwide aquaculture production of abalone has greatly increased in the last decades. Selective breeding has been initiated to improve growth in major abalone species. In France, the abalone production was initiated 30 years ago and struggles to develop. This CIFRE PhD co-financed by France Haliotis, the only French hatchery in activity, was initiated to study early domestication process of farmed abalone population and to provide knowledge required to design a selective breeding program adapted to this company. Pathology of cultured paua Haliotis iris infected with a novel haplosporidian parasite, with some observations on the course of disease ArchiMer

Mortalities among juvenile paua Haliotis iris Martyn 1784 in a commercial culture facility were reported in April 2000. Histology of moribund paua showed heavy systemic infections of a uni- to multi-nucleate stage of a novel organism later confirmed by transmission electron microscopy (TEM) and molecular studies to be a haplosporidian. Multinucleate plasmodia up to 25 μm diameter with up to 17 nuclei were detectable in wet preparations of hemolymph from heavily infected paua. The presence of the haplosporidian in the affected facility was associated with mortalities of slow growing 'runt' paua during the summer months. The development of a loop-mediated isothermal amplification assay for rapid and sensitive detection of abalone herpesvirus DNA ArchiMer

A loop-mediated isothermal amplification (LAMP) assay was developed for the detection of abalone herpesvirus DNA. Two pairs of primers were designed, based on the sequence of the DNA polymerase gene of abalone herpesvirus. The reaction temperature and time were optimized to 63 °C and 60 min, respectively. LAMP amplicons were analyzed by 2% agarose gel electrophoresis or by visual inspection of a colour change emitted by fluorescent dye. The method developed was specific for the detection of abalone herpesvirus, without cross-reactions with other tested herpesviruses including ostreid herpesvirus 1 (OsHV-1), European eel herpesvirus, koi herpesvirus (KHV) and an avian herpesvirus. The growth of the abalone Haliotis Tuberculata L. in Bay of Brest ArchiMer

The growth of H. tuberculata L. was studied by analysing the distribution of the growth checks on the shell of 640 individuals. The average lengths of the seven first rings including the first one appear to fit weIl to the Von Bertalanffy model. The value of the growth parameter K = 0,33 is rather similar to those cited in Litterature for the same species but the average attainable size L%= 97,5 seems to be low in this area.

Transcriptome based SNP discovery and validation for parentage assignment in hatchery progeny of the European abalone Haliotis tuberculata ArchiMer

Selective breeding strategies require pedigree information over generations, but many species produced in aquaculture are too small to be physically tagged at early stages. Consequently, maintaining a sufficient number of separate families is often needed but costly and logistically difficult. Alternatively, parentage assignment can be obtained using DNA markers. Although a wide range of viruses have been reported in marine molluscs, most of these reports rely on ultrastructural examination and few of these viruses have been fully characterized. The lack of marine mollusc cell lines restricts virus isolation capacities and subsequent characterization works.
